Ok I represent a dude whose wife left him for another woman. My client has 2 kids with his ex and the ex's lesbian lover has two kids with her now ex husband. The two women are now married and live with all 4 children.

My client was contacted by the ex-husband of his ex-wifes wife who is now in the midst of post-judgment litigation with his ex-wife.

Can i represent both dudes against their lesbian ex-wives?

Not legal advice, what follows is all made up bullshit I'm writing out while taking a dump,

1) assuming this is a waivable conflict, make sure you get these dudes' informed consent and have em waive in writing

2) the fact their ex -wives are now married makes me nervous about any monetary claims these dudes may have stemming from their respective divorces. They may have materially adverse interest re any of the dykes' property.

In any case, unless these ladies or dudes are loaded, this is probably not worth your time.

Yes you can. There is no conflict raised by this issue, at least under New York law which mirrors the MPC. You'd be representing two former husbands against their former wives. There would only be a conflict if you were representing or had previously represented one of the former wives in which case you would be representing someone materially adverse to the other party in the same or a substantially related matter. There are some other provisions that might be triggered if we toyed with the hypothetical but I don't see the point.
