A third party will need to become popular for long enough to get First Past The Post replaced with an alternative election system in which vote% better matches legislative seat %. (Ie proportional representation.)
Most plausible path to this happening would start with these reforms being put in place at the local level.

The two party system has been remarkably resilient over the course of American history. The Federalists collapsed and briefly there was one party rule in the USA, which lasted for less than a decade before a new party emerged. Same with the Whigs collapsing - the GOP appeared in just a few years and won the POTUSy in their first five years.
Perhaps demographic change will kill the GOP, but something else will replace it.
