At some point, there aren't many doors open and possibilities left in your life. In your 20s, you can find young love, start fresh in an industry, be somebody. By 40, you're very likely on the path you're on, and if you haven't already made it in whatever you want to make it in, you're probably not going to.
Two saving graces are (1) having kids, which resets the clock insofar as they can still make it and be and do anything, and (2) making enough money to get out and the wage cage and at least chill for a bit.

The vast majority of people have a kid by 40--even in our declining birth rate techno-dystopia.
One definitely can, in theory, turn the ship around in his/her 40s and above. Some people have. E.g., Rodney Dangerfield found little success as a comic in his 20s, he quit to get a regular job in his 30s, and he returned to comedy in his 40s, becoming one of the biggest comics of his era and a legend.
The problems are more that (a) very few people who haven't already found success or put themselves on the path to success by 40 are going to have the resources, will power, energy, capacity, etc. to do so after 40 and (b) even if the factors in (a) weren't a problem, a lot of doors still remain closed, by choice or otherwise.

Entertainment--40smos aren't "up" on what's cool, no one wants to see a 40 year old struggling to make it and find his footing, youthful energy and creativity are gone.
Anything that requires being away from family and starting out with little pay. E.g., I'm a reasonably competent guitarist and I could find a band to tour with (but see above about no one wanting to see a 40 year old entertainer who isn't at the top of the craft or who isn't already famous). However, I don't want to be away from my kids, and I want to make money so they can live a good life.
Breaking into a new industry that requires starting on the ground floor. That's cool and fine when you're in your 20s. Would you want to be a 43 year old first year associate? Junior programmer?
